In the field of  “Chromatin-based mechanisms of genome maintenance” the research group of Dr. Vassilis Roukos offers the following PhD project:

Genome-wide analysis of double-strand break-repair dynamics


Experimental limitations in quantifying repair efficiency throughout the genome have previously precluded the assessment of DSB repair efficiency in the context of the genome architecture. We are in the process of developing a system that allows the induction and repair of DSBs in a time-controlled and genome-wide manner in mammalian cells. By using established break-quantification methods combined with high-throughput sequencing, this project aims at the evaluation of the repair efficiency across the genome and the correlation in silico with previously reported topological maps of distinct chromatin domains. The PhD student will establish the experimental system, perform the quantification experiments and the acquired sequencing data will be analyzed in collaboration with computational scientists. Therefore, an interest in bioinformatics is beneficial for prospective candidates. These studies will for the first time systematically probe DSB-repair in the context of genome architecture.
