The role of Gadd45 in mouse development

Description:

Our studies have also indicated a prominent role for Gadd45 in mouse development. Gadd45g homozygous mutant mice are sex reversed! Male mice transform into females very early during embryonic development. The reason is that the sex determination gene SRY is misregulated in Gadd45g mutant mice (Gierl et al., 2013). These studies have raised interesting new questions regardin the function of Gadd45 family genes in mouse. What are the Gadd45 target genes in mice? How do they contribute to the observed phenotypes in Gadd45 mutants? How is Gadd45 recruited to its target genes? Does Gadd45 demethylation in mouse involve oxidized cytosine intermediates?  In this project we will study the role of Gadd45 using mouse as model system. Mutant mice will be phenotypically analyzed by marker gene analysis using qPCR, immunostaining, and in situ hybridization. We will carry out global expression profiling by microarrays and/or RNA-sequencing in combination with global methylation profiling using MeDIP and Gadd45 ChIP-Seq to identify Gadd45 target genes in embryonic differentiation.
