In the field of  “Mathematical modelling of single-cell responses” the research group of Dr. Stefan Legewie offers the following PhD project:

Harnessing the multivariate nature of single-cell data to derive meaningful models of cellular processes


The predictive power of mechanistic mathematical models is often limited due to sloppiness and non-identifiability of kinetic parameter values. One problem is that the models are typically calibrated based on population average data which hides information about nonlinear phenomena and correlations in single-cells. To overcome this limitation, we describe cohorts of individual cells in cellular ensemble models, and simultaneously fit them to single-cell and cell population data for calibration [1]. This approach accommodates that comprehensive single-cell measurements are typically restricted to certain signalling intermediates, while less informative, population-based assays can be done on a higher throughput. In this PhD project, the cellular ensemble modeling approach should be further developed in close collaboration with experimental partners (Silke Hauf, Virginia Tech) to allow´for a quantitative description of a network controlling chromosome segregation during cell division.
