In the field of “Super-Resolution Microscopy of Nuclear Nanostructure” the research group of Professor Christoph Cremer offers the following PhD project:

Imaging the epigenetic Landscape of Nuclear Genome Nanostructure in Three


We have designed and realized various SRM systems which allow us to study nuclear structure in three dimensions (3D), both in fixed and in live cells. In the biologically oriented Ph.D. thesis proposed here, the goal is to study quantitatively the 3D distribution of chromatin in different cell types down to the single molecule resolution level under different conditions of environmental stress, and to correlate this 3D distribution with epigenetic markers for gene activity, replication, splicing and repair. In collaborative projects with other laboratories, the spatial 3D nanoscale data obtained are planned to be compared with results based on sequencing approaches, such as chromosome conformation capture (Hi-C), as well as numerical Biocomputing predictions of chromatin structure. The results will contribute substantially to the understanding of the molecular mechanisms involved in the fine tuning of gene expression in normal and aberrant cells, as well as in cellular reprogramming.
