PhD position in Telomere Biology

In the field of “Crosstalk between DNA damage checkpoints and nutrient signalling” the research group of Dr Brian Luke offers the following PhD project:

Understanding the crosstalk between checkpoint and nutrients

With regards to the non-coding RNA TERRA, we are using yeast and human cells to understand how RNA-DNA hybrids (R-loops) are regulated at telomeres throughout the cell cycle and also in cells that have escaped senescence (similar to cancer cells using the ALT mechanism). We are employing live cell microscopy, chromatin immunoprecipitation, Southern blotting and many other molecular, genetic and biochemical assays to address these questions. We believe that a better understanding of TERRA regulation could eventually provide a specific means to target ALT cancers.

For the checkpoint adaptation projects, we are now focusing on chemotherapeutic drugs and addressing how the manipulation of the checkpoint can influence the efficacy of such drugs. Here also we are employing both yeast and human cells to interrogate these questions. We want to understand how the molecular signaling between metabolic pathways and the checkpoint is organized. Our preliminary data suggests that this interaction has an important influence on whether or not cells become resistant to chemotherapeutics.
