PhD project: The role of demethylation genes in Xenopus embryonic development

In the field of  “DNA demethylation” the research group of Prof. Christof Niehrs offers the following PhD project: The role of demethylation genes in Xenopus embryonic development

Description:

To unravel the biological role played by genes in active DNA demethylation we are using the Xenopus frog system. Xenopus embryos are a powerful model system to study embryonic patterning and differentiation. Tools such as mRNA overexpression and antisense Morpholino injection yield facile access to functional analysis of gene function. These studies have raised interesting questions: What are the demethylation target genes in Xenopus? What is the role of demethylating genes such as Gadd45 or Neil DNA glycosylases? Does demethylation in Xenopus involve oxidised cytosine intermediates? We will combine genome-wide expression and methylation analysis. Global expression profiling by microarrays and/or RNA-sequencing in combination with global methylation profiling using MeDIP and ChIP-Seq will lead to the identification of direct demethylation target genes in this differentiation process.
