The endocrine acting protein fibroblast growth factor 21 (FGF21) is an important multiorgan metabolic regulator promoting health span and increasing lifespan. FGF21 was shown to improve glucose and lipid metabolism and reduce obesity in rodent models and non-human primates. However, despite the unequivocal beneficial metabolic effects of FGF21 there are still uncertainties and controversies concerning its physiology especially regarding the exact cellular targets and intracellular mechanisms of FGF21 signaling which is still poorly understood and further pre-clinical needs to be conducted. For these experiments, FGF21 needs to be produced via transient expression in Nicotiana benthamiana (N. benthamiana) and purified from the plant biomass.

Activities and responsibilities

The student accepted for the project described above conduct the transient expression of recombinant FGF21 in N. benthamiana plants, including the cultivation of Agrobacterium tumefaciens, harboring the FGF21 expression vector, infiltration of whole plants, biomass harvesting and FGF21 purification. The isolated protein well be analyzed via Western blot and ELISA.

Qualification profile

The student successfully applying for this project is skilled in written and spoken English to familiarize her/himself with the relevant protocols and to fluently communicate within the international atmosphere of the Fraunhofer IME. (S)He has good basic knowledge in plant biochemistry and bioengineering and is willing to gain more insights into the applied biotechnology and plant-made pharmaceuticals.

Working conditions
The working time will be approximately 6 - 9 hours per week.
The position is initially limited to several months.
