Master Thesis Position - Screening, Identification and Characterization of Fucoidanase

Development of the biocatalysis-based industry "white industry" need to screen for new enzyme, which can hydrolysis the available biomass. Based on their chemical composition, macroalgae are an attractive third generation feedstock for oleo-chemicals and surfactants. Macroalgae are available in large quantities and can be easily collected from coastal regions, e.g. from the North and Baltic Sea. Fucoidan, is a sulfated polysaccharides, attached a considerable interesting form the industry partners. Screening for fucoidanase one of important task for the utilization of biomass. In this project, different strains (4-5 strains) from environmental samples have been detected to have a fucoidanase activity. Metagenomic (λ-ZAP gene lib.) approach will be used to screening for this enzyme activity. Proteomic identification and biochemical characterization on detected enzymes will carried on. The gene of protein will be sequenced from the original strain in order to get the whole operon. In parallel, screening for new strains from environmental samples will be also required.
Responsibility and Tasks:
  • Building λ-ZAP library.
  • Screening for fucoidanase activity from the gene lib.
  • Screening for related enzyme activity from environmental samples.
  • Charactrization of detected protein

  • HPLC, TLC, GC-FID, GC-MS, TOF-MALDI-MS and plate reader…
  • Protein characterization techniques SDS-page, IEF-Page, FPLC…
  • Screening techniques: λ-ZAP library, Auto-picker…
  • Screening using RBB or CPH screening methods.
  • Student from biology, biotechnology, biochemistry or equivalent courses are sutable candidate. Hrd working, High team spirit and communication skills, as well as an independent operation are important charachteristics. Experience in the areas of responsibility listed above are an advantage.
