„Optimization of process parameters for the perfusion-based cultivation of an animal cell line using a tangential flow filtration (TFF) system“
Cell cultivation in perfusion mode consists in the continuous addition of nutrients at the same flow rate that exhausted medium enriched with the product of interest is extracted from the bioreactor. During perfusion, cells are contained within the bioreactor using a retention system, which enables to reach very high cell and product concentrations. The TFF-based retention system has proved its suitability to reach very high cell concentrations. However, operation parameters for tangential filtration should be set according to each cell line.
Within this project, optimal filtration parameters such as transmembrane pressure, flow rate and linear velocity, using a TFF-based perfusion system, should be established to minimize shear stress on an animal cell line. Optimal conditions should be used in further cultivations to achieve high cell concentrations.
